二. ALPHA COMPLEMENTATION/MOLECULAR BIOLOGY 分子生物学:
Iacz基因作为一种通讯基因在微生物分子生物学中被频繁使用,Iacz编译一种能够水解底物X-β-D-gal,形成一种靛蓝染料沉淀,很多常见的无性繁殖的带菌者在DNA片断插入复制时都有Iacz基因被表达DNA片断插入Iacz片断时,通常阻碍Iacz片断工作,只有一小部分Iacz基因片断用于保持质粒体大小。Iacz基因超过3000个碱基,而且大部分复制质粒体比它还小,这些小片断叫作α区,位于Iacz基因起始端,α区基因可以通过基因互补来控制β葡萄糖苷酶的活性,所以细胞中Iacz基因被检测出α区变异。β葡萄糖苷酶的α片断和形成活性β葡萄糖苷酶的变异的α片断相第影响,那此被无性繁殖的带菌都插入的细菌可以被真实的检出。蓝色菌落表示α互补,白色或无色就可能是有克隆片断插入Iacz基因。
IPTG通过阻断Iac增强了β-葡(萄)糖苷酶的活性。
培养基和生长条件:LB琼脂平板,加入100mg/ml X-β-D-gal, 100mg/ml IPTG 37℃培养24小时。
LacZ reporter gene system
The lacZ gene is frequently used in molecular biology as a reporter gene. LacZ encodes the enzyme beta-Galactosidase which can hydrolyze the substrate X-β-D-gal (5-Bromo-4-chloro-3-indoxyl-beta-D-galactopyranoside, B-7150) to form a precipitating blue indigo dye. Many popular cloninq vectors have part of the lacZ gene to indicate when DNA fragments are cloned (inserted) into them. The DNA fragment, tLen inserted into the lacZ fragment, usually inactivates (blocks) the lacZ gene fragment from working. Qnly a Small fragment of the lacZ gene is used in order to keep the size of the plasmid small. (The IacZ gene is over 3000 base pairs long, and most cloninq plasmids are smaller than that.) This small fragment is called the alpha region and is located in the beginning ot the lacZ gene. The alpha portion of the gene can result in beta-Galactosidase enzyme activity by a process called “intragenic complemen-tation” whereby the rest of the lacZ gene is provided in the cell by an alpha mutant of the lacZ gene.The alpha fragment of the beta-Galactosidase enzyme interacts with the complementary portion of the enzyme provided by the alpha mutant to form an active beta-Galactosidase enzyme. Therefore,bacterial colonies can be visually screened to determine which colonies have the cloning vector inserted. Blue colonies show alpha complemen-tation, While white or colorless colonies probably have the cloned fragment inserted within the lacZ gene.

七. FOOD MICROBIOLOG 食品微生物学:
E. coli O157:H7(病原性大肠杆菌)是一种危险的损伤E. coli 时常存在于汉堡,鸡肉和其他肉食品中。根据USDA大概10000人每年因为不熟的包含着E. coli O157:H7的肉制品而染病,这个危险的有机体能够引起很多疾病甚至死亡。
培养基和生长条件:
麦克拉肯山梨(糖)醇 琼脂平盘含100ug/ml X-glcUA·Na(BClG), 35℃过液,大部分的含β-葡(萄)糖苷酸酶的E. coli会和酶底物X-glcUA·Na反应成为蓝色菌落,没有β-葡(萄)糖苷酸酶的大肠杆菌为无色状态。
Testing for E. coli O157:H7
E. coli O157:H7 is a dangerous strain of E.coli which is sometimes present in hamburger, chicken, and other meat products, According to the USDA, as many as 10,000 people each year become sick from eating undercooked meat containing E. coli O157:H7. This dangerous organism can cause severe illness and even death.
Culture media and growth conditions
MacConkey Sorbitol Agar Plates with 100 micrograms per ml of X-glcUA·Na (also known as BClG). Plates were incubated overnight at 35℃. Most E. coli contain the enzyme beta-Glucuronidase, which reacts with the enzyme substrate X-glcUA·Na to produce the deep blue seen in the colonies. E. coli O157:H7 is a dangerous strain of E.coli which is sometimes present in hamburger, chicken, and other meat products, According to the USDA, as many as 10,000 people each year become sick from eating undercooked meat containing E. coli O157:H7. This dangerous organism can cause severe illness and even death.
Culture media and growth conditions
MacConkey Sorbitol Agar Plates with 100 micrograms per ml of X-glcUA·Na (also known as BClG). Plates were incubated overnight at 35℃. Most E. coli contain the enzyme beta-Glucuronidase, which reacts with the enzyme substrate X-glcUA·Na to produce the deep blue seen in the colonies. Enzyme inducers and enhancers:
SEEBIO offers a number of reagents which either induce or enhance several important enzyme/substrate reactions. Our most recent and very effective inducer is Isopropyl-beta-D-thioglucuronic acid, sodium salt (I-8350).
Isopropyl-beta-D-thioglucuronic acid sodium salt is a “gratuitous” inducer, i.e. it is not hydrolyzed by the enzyme, and enhances very effectively the sensitivity of your beta-Glucuronidase test in E. coli by increasing the concentration of the enzyme.

C-390是一种抗菌剂加入到琼脂培养基是,可以有效地将假单胞铜绿菌从其他细菌分离出来。
使用情况:多种细菌最小抑制浓度实验的结果表示,C-390在25ug/ul或更少浓度时,抑制微生物(除假单)的生长,然而MICS包含从50到大于100ug/ml的假单胞菌铜绿菌,因此C-390被评价为是在假单胞菌琼脂F中的最有潜力的选择性选择试剂,修复试验在53株假单胞菌铜绿菌的存在下进行,结果和在其他培养基作对比,显示假单胞菌琼脂F含30ug/ml C-390抑制性小而选择性大,加入C-390的假单胞菌琼脂F,有选择性和差异性,大量的证据同样告诉我们,C-390可以被加入到其他基础培养基中,得到同样的结果,文献著作权属SEEBIO所有。

Isolation of Pseudomonas aeruginosa
C-390 is an antibacterial agent which is useful for the selective isolation of Pseudononas aeruginosa from other bacterial species when added to an agar medium.
Patent: U.S. 4,263,398(4/21/81)
Use: Results of minimal inhibitory concentration tests with a diversity of bacterial strains showed that C-390 inhibited the growth of all microorganisms tested (other than Pseudononas aeruginosa) at 25μg/μl or less, whereas MICs obtained for Pseudononas aeruginosa ranged from 50 to >100μg/ml. Therefore, C-390 was evaluated as a potential selective agent for Pseudononas aeruginosa in pseudomonas agar F.Recovery tests were conducted on this medium with 53 strains of Pseudononas aeruginosa, and the results were compared to those obtained in similar tests on commercially available selective media, i.e. pseudomonas isolation agar and Psersosel agar. The results of these comparisons indicated that pseubomonas agar F with C-390(30μg/ml) was significantly less inhibitory than Pseudosel agar and pseudomonas isolation agar and more selective than pseudomonas isolation agar. The incorporation of C-390 in pseudomonas agar F also provided a medium that was both selective and differential. Preliminary evidence also suggested that C-390 may be added to other basal media with comparable results. Literature is available from SEEBIO.

Isolation of Pseudomonas aeruginosa
C-390 is an antibacterial agent which is useful for the selective isolation of Pseudononas aeruginosa from other bacterial species when added to an agar medium.
Patent: U.S. 4,263,398(4/21/81)
Use: Results of minimal inhibitory concentration tests with a diversity of bacterial strains showed that C-390 inhibited the growth of all microorganisms tested (other than Pseudononas aeruginosa) at 25μg/μl or less, whereas MICs obtained for Pseudononas aeruginosa ranged from 50 to >100μg/ml. Therefore, C-390 was evaluated as a potential selective agent for Pseudononas aeruginosa in pseudomonas agar F.Recovery tests were conducted on this medium with 53 strains of Pseudononas aeruginosa, and the results were compared to those obtained in similar tests on commercially available selective media, i.e. pseudomonas isolation agar and Psersosel agar. The results of these comparisons indicated that pseubomonas agar F with C-390(30μg/ml) was significantly less inhibitory than Pseudosel agar and pseudomonas isolation agar and more selective than pseudomonas isolation agar. The incorporation of C-390 in pseudomonas agar F also provided a medium that was both selective and differential. Preliminary evidence also suggested that C-390 may be added to other basal media with comparable results. Literature is available from SEEBIO.
