Isopropyl –β-D-thiogalactopyranoside (IPTG) is a potent inducer of the lac operon which irreversibly binds the lac repressor. The lac operon is a commonly used genetic element in transcription research for the detection of gene insertion. IPTG binds to a specific site on the lac repressor causing a conformational change which results in its dissociation from the lac operon. While IPTG structurally resembles the physiological inducer 1,6-allolactose, IPTG overcomes β-allolactose's susceptibility to degradation by the β-galactosidase enzyme. Since IPTG is not metabolized it permits the separation of the physiological function of lactose as a carbon source from its function in the regulation of lac gene expression. These properties make IPTG ideal for use in industrial applications for the large scale production of expressed compounds.
IPTG is often used in conjunction with a β-galactosidase substrate such as X-GAL (5-Bromo-4-Chloro-3-Indolyl-beta-D-Galactopyranoside CAS: 7240-90-6) or BLUO-GAL (5-Bromo-3-Indolyl-beta-D-Galactopyranoside CAS: 97753-82-7) for determination of the lac phenotype in recombinant organisms.
