寡核苷酸,是一类20个左右碱基的短链核苷酸的总称(包括脱氧核糖核酸DNA或核糖核酸RNA内的核苷酸)。寡核苷酸可以很容易地和它们的互补链结合,所以常用来作为探针确定DNA或RNA的结构;而其作为药物候选应用的研究则始于大约30年前,包括了反义寡核苷酸(ASOs),核酸适配体(apatmers)及近15年来对各类siRNAs的研究。这期间,医药工业界开展了大量的临床试验。截至2017年5月底,共有6类寡核苷酸类药物获得美国FDA批准上市应用,其中属于反义寡核苷酸的药物有四种,同时有大量药物处于不同阶段的临床研究中。[查看]

Science:重磅!揭示一种新的DNA损伤修复机制
